Bipolar Disorder01:30

Bipolar Disorder

Bipolar disorder is a chronic mental health condition marked by significant mood fluctuations, including episodes of mania and depression. Elevated energy levels, heightened mood or irritability, impulsive behavior, reduced sleep needs, rapid speech, racing thoughts, inflated self-esteem, and distractibility characterize mania. Individuals with bipolar disorder often alternate between depressive and manic states, with periods of emotional stability lasting an average of six months to a year.
Human Genetics01:28

Human Genetics

Human genetics provides a profound framework for understanding the interplay between genetic predispositions and human psychology. At the heart of this discipline lies the study of how genes influence physical traits, behaviors, and susceptibility to diseases. Each person carries a unique genetic code that subtly or significantly shapes their psychological and behavioral landscape.
The complex relationship between genetics and psychology is observable through common biological components such...

Summary
This summary is machine-generated.

Epigenetic changes in the HCG9 gene are linked to bipolar disorder (BPD). DNA methylation differences were found across brain, blood, and sperm, suggesting a potential causal role for HCG9 in BPD.

Area of Science:

  • Molecular Psychiatry
  • Epigenetics
  • Neuroscience

Background:

  • Epigenetic modifications, including DNA methylation, are increasingly recognized as crucial in understanding human diseases.
  • Previous epigenome-wide scans identified DNA methylation differences in post-mortem brain samples associated with major psychosis.

Purpose of the Study:

  • To fine-map DNA methylation differences at the human leukocyte antigen (HLA) complex group 9 gene (HCG9) in bipolar disorder (BPD).
  • To investigate the association of HCG9 DNA methylation with BPD across multiple tissue types.

Main Methods:

  • Utilized sodium bisulfite conversion and pyrosequencing to analyze 28 CpGs in the HCG9 gene region.
  • Examined 1402 DNA samples from post-mortem brains, peripheral blood cells, and germline (sperm) from BPD patients and controls.
  • Analyzed approximately 40,000 CpGs to explore relationships between DNA methylation, age, medication, and DNA sequence variation (rs1128306).

Main Results:

  • Lower DNA methylation in an extended HCG9 region was observed in brain tissue from BPD patients compared to controls (P=0.026).
  • Independent effects of DNA methylation in white blood cells (OR=1.08, P=0.0077) and overall samples (OR=1.24, P=0.0011) were identified for BPD prediction.
  • Sperm DNA showed a significant association (P=0.018) with BPD in a region consistent with epigenetic changes in brain and blood.

Conclusions:

  • Consistent, multi-tissue epigenetic differences at HCG9 suggest a causal association with bipolar disorder.
  • DNA methylation patterns at HCG9 may serve as a biomarker for BPD prediction, with potential implications for understanding disease etiology.
