Area of Science:

  • Aging biology
  • Model organism research
  • Dietary interventions

Background:

  • Dietary restriction (DR), or caloric restriction (CR), is known to increase lifespan and delay age-related diseases in mammals.
  • The nematode Caenorhabditis elegans is a key model organism for aging research.
  • Existing DR studies in C. elegans often use liquid media, differing from standard solid-plate genetic studies.

Purpose of the Study:

  • To develop and present a practical dietary restriction (DR) protocol for the nematode C. elegans.
  • To enable DR studies on standard solid agar plates, aligning with common genetic aging research methods.

Main Methods:

  • Development of a DR protocol using standard Nematode Growth Medium (NGM) agar plates.
  • Utilized killed bacteria as a food source within the solid NGM medium.

Main Results:

  • Successfully established a dietary restriction (DR) method on solid NGM plates for C. elegans.
  • This protocol facilitates DR studies in a manner consistent with standard genetic aging research.

Conclusions:

  • The presented solid-plate DR protocol offers a more accessible and standardized approach for aging research in C. elegans.
  • This method bridges the gap between liquid-based DR studies and solid-plate genetic studies, potentially advancing aging research.
