Summary

Researchers developed the first sunflower TILLING resource, a powerful tool for identifying ethyl methanesulfonate (EMS)-induced mutations. This resource aids in understanding gene function in sunflower, a vital oilseed crop.

Area of Science:

  • Plant genetics and genomics
  • Crop improvement
  • Mutagenesis

Background:

  • Sunflower (Helianthus annuus L.) is a key global oilseed crop with extensive genetic and genomic research.
  • Classical mutagenesis has been used, but a dedicated TILLING resource was lacking.

Purpose of the Study:

  • To establish the first TILLING resource for cultivated sunflower.
  • To optimize ethyl methanesulfonate (EMS) treatment for mutation induction.
  • To assess the efficiency of TILLING in identifying induced mutations.

Main Methods:

  • 'Kill curve' analysis to determine optimal EMS dosage and exposure time (0.7% EMS for 6 h).
  • Generation of an M2 population of 3,651 fertile plants.
  • TILLING procedure using CelI-nuclease mismatch cleavage assay and DNA pooling.

  • Pilot screening of 1,152 M2 lines across four genes.
  • Main Results:

    • An optimal EMS treatment (0.7% for 6 h) was identified, yielding 3,651 M2 plants with 4.79% showing aberrant phenotypes.
    • Microsatellite analysis confirmed genetic uniformity of the mutant lines.
    • Pilot screening identified 9 mutant lines, yielding a mutation frequency of 1/475 kb.

    Conclusions:

    • A TILLING population for high-throughput identification of EMS-induced point mutations in sunflower has been successfully created.
    • This resource is a valuable tool for advancing gene function studies in sunflower.
