Enterobacterial Repetitive Intergenic Consensus PCR (ERIC-PCR) effectively differentiates Corynebacterium pseudotuberculosis strains from sheep. This molecular typing tool shows high discriminatory power for identifying disease-causing agents in livestock.
Area of Science:
- Veterinary Microbiology
- Molecular Epidemiology
- Bacterial Typing
Background:
- Caseous lymphadenitis is a significant infectious disease in sheep and goats.
- Corynebacterium pseudotuberculosis causes caseous lymphadenitis, presenting as lymph node abscesses.
- Existing typing methods for C. pseudotuberculosis are often inadequate.
Purpose of the Study:
- To evaluate Enterobacterial Repetitive Intergenic Consensus PCR (ERIC-PCR) for molecular typing of C. pseudotuberculosis.
- To assess the discriminatory potential of ERIC-PCR for isolates from sheep in Brazil.
Main Methods:
- One hundred twenty-seven C. pseudotuberculosis isolates were collected from sheep lesions in Minas Gerais, Brazil.
- Molecular fingerprinting was performed using Random Amplified Polymorphic DNA (RAPD) with ERIC primers (ERIC-1R, ERIC-2, and ERIC-1R+ERIC-2).
- Genotyping and discriminatory power were assessed using the Hunter-Gaston Discrimination Index (HGDI).
Main Results:
- ERIC-PCR generated 17 to 21 distinct genotypes among the isolates.
- The HGDI values ranged from 0.69 to 0.87, indicating good discriminatory power.
- High correlation (W Kendall = 0.91) was observed among the different ERIC-PCR primer combinations.
Conclusions:
- ERIC-PCR is a valuable and effective tool for molecular typing of C. pseudotuberculosis in sheep.
- This method offers advantages over existing DNA-based typing techniques for epidemiological studies.
- The findings support the use of ERIC-PCR for discriminating C. pseudotuberculosis isolates in disease outbreak investigations.
