Insights
Circulating CD90(+) CD73(+) CD45(-) cells are more prevalent in cirrhosis patients and exhibit enhanced in vitro proliferation, suggesting a role in liver disease progression.
Area of Science:
- Cell biology
- Immunology
- Hepatology
Background:
- Liver cirrhosis is a severe condition with limited treatment options.
- Understanding cellular mechanisms in cirrhosis is crucial for developing new therapies.
Purpose of the Study:
- To identify and characterize circulating CD90(+) CD73(+) CD45(-) cells in liver cirrhosis.
- To evaluate the in vitro proliferative capacity of these cells.
Main Methods:
- Peripheral blood mononuclear cells were isolated from cirrhosis patients and healthy controls.
- Cells were cultured, and fibroblast-like cells were analyzed for CD90, CD73, and CD45 expression using flow cytometry and immunocytochemistry.
- Colony-forming efficiency (CFE) was assessed to evaluate proliferative potential.
Main Results:
- Cirrhosis patients exhibited a significantly higher percentage of CD90(+) CD73(+) CD45(-) cells compared to healthy volunteers.
- These cells demonstrated a tenfold increase in CFE, indicating enhanced proliferation.
- Flow cytometry and immunocytochemistry confirmed the expression of CD90(+) CD73(+) markers on proliferating cells.
Conclusions:
- Circulating CD90(+) CD73(+) CD45(-) cells are present in higher numbers in liver cirrhosis patients.
- These cells possess a significantly elevated capacity for in vitro proliferation.
- The findings suggest these cells may play a role in the pathophysiology of liver cirrhosis.
Aim:
To identify circulating CD90(+) CD73(+) CD45(-) cells and evaluate their in vitro proliferating abilities.
Methods:
Patients with cirrhosis (n = 43), and healthy volunteers (n = 40) were recruited to the study. Mononuclear cells were isolated and cultured from the peripheral blood of controls and cirrhosis patients. Fibroblast-like cells that appeared in cultures were analyzed for morphological features, enumerated by flow cytometry and confirmed by immunocytochemistry (ICC). Colony forming efficiency (CFE) of these cells was assessed and expressed as a percentage.
Results:
In comparison to healthy volunteers, cells obtained from cirrhotic patients showed a significant increase (P < 0.001) in the percentage of CD90(+) CD73(+) CD45(-) cells in culture. Cultured cells also showed 10 fold increases in CFE. Flow cytometry and ICC confirmed that the proliferating cells expressed CD90(+) CD73(+) in the cultures from cirrhosis patients.
Conclusion:
These results indicate the presence of circulating CD90(+) CD73(+) CD45(-) cells in patients with liver cirrhosis that have the potential to proliferate at a higher rate.
