Adaptation of poliomyelitis virus type I (Mahoney strain) to the CNS of cotton rats which at the time of inoculation had been temporarily immunosuppressed with cyclophosphane is described. In the course of passages the virus properties were observed to change. The virus became pathogenic for cotton rats with normal immunologic responsiveness by the intracerebral, intraperitoneal and subcutaneous routes, as well as for white mice intracerebrally. Virions pathogenic for cotton rats were predominant in the adapted strain population.
