Proteomics

A proteome is the entire set of proteins that a cell type produces. We can study proteomes using the knowledge of genomes because genes code for mRNAs, and the mRNAs encode proteins. Although mRNA analysis is a step in the right direction, not all mRNAs are translated into proteins.
Proteomics is the study of proteomes' function. It involves the large-scale systematic study of the proteome to denote the protein complement expressed by a genome. Scientist Mark Wilkins coined the term proteomics...
Peptide Identification Using Tandem Mass Spectrometry01:33

Peptide Identification Using Tandem Mass Spectrometry

Tandem mass spectrometry, also known as MS/MS or MS2, is an analytical technique that employs two mass analyzers. Essentially it is a series of mass spectrometers that helps isolate a particular biomolecule and then helps study its chemical properties.
This technique helps gather information regarding the protein from which the peptide was obtained and to study the peptides’ amino acid sequence. Identifying peptides from a complex mixture is an important component of the growing field of...

Area of Science:

  • Biochemistry
  • Molecular Biology
  • Proteomics

Background:

  • Lack of sensitive and specific methods for parallel protein biomarker measurement in large sample sets.
  • Need for advanced techniques in biomarker discovery and validation.

Purpose of the Study:

  • To present and validate a novel multiplex proximity ligation assay, ProteinSeq, for enhanced protein biomarker analysis.
  • To identify potential diagnostic biomarkers for cardiovascular disease (CVD).

Main Methods:

  • Development of a multiplex proximity ligation assay (ProteinSeq) with readout via real-time PCR or DNA sequencing.
  • Simultaneous analysis of 35 proteins in small plasma volumes (5 µl).
  • Comparison of sensitivity and specificity against conventional sandwich assays.

Main Results:

  • ProteinSeq demonstrated improved sensitivity over conventional sandwich assays for multiplex protein analysis.
  • Minimal background increase observed with multiplexing, suggesting scalability.
  • Identification of P-selectin, Cystatin-B, and Kallikrein-6 as putative CVD diagnostic biomarkers in plasma samples.

Conclusions:

  • ProteinSeq is a promising technology for high-throughput screening and validation of protein biomarkers.
  • The identified biomarkers require further validation in larger patient cohorts.
  • ProteinSeq offers a valuable platform for biobank sample analysis and clinical diagnostics.
