Super-resolution Fluorescence Microscopy01:37

Super-resolution Fluorescence Microscopy

Super-resolution fluorescence microscopy (SRFM) provides a better resolution than conventional fluorescence microscopy by reducing the point spread function (PSF). PSF is the light intensity distribution from a point that causes it to appear blurred. Due to PSF, each fluorescing point appears bigger than its actual size, and it is the PSF interference of nearby fluorophores that causes the blurred image. Various approaches to achieving higher resolution through SRFM have recently been developed.

Imaging Biological Samples with Optical Microscopy

Optical microscopy uses optic principles to provide detailed images of samples. Antonie van Leeuwenhoek designed the first compound optical microscope in the 17th century to visualize blood cells, bacteria, and yeast cells. In 1830, Joseph Jackson Lister created an essentially modern light microscope. The 20th century saw the development of microscopes with enhanced magnification and resolution.
In optical microscopy, the specimen to be viewed is placed on a glass slide and clipped on the stage...

Area of Science:

  • Biomedical optics
  • Photoacoustic imaging
  • Microscopy

Background:

  • Optical-resolution photoacoustic microscopy (OR-PAM) offers high-resolution imaging capabilities.
  • Existing OR-PAM systems are often large, stationary, and less suitable for clinical settings.
  • There is a need for more portable and user-friendly OR-PAM systems for diverse applications.

Purpose of the Study:

  • To demonstrate a new generation of compact, portable OR-PAM systems.
  • To evaluate the imaging performance and potential clinical applicability of the developed OR-PAM system.
  • To showcase the system's ability to visualize microvasculature in vivo.

Main Methods:

  • Development of a portable OR-PAM probe (4 cm x 6 cm, ~500 g) utilizing fast scanning mirrors and an image guide with 30,000 fiber pixels.
  • Integration of a refocusing lens and a unique probe design for enhanced performance.
  • Phantom studies to assess resolution and in vivo imaging of mouse ear microvasculature.

Main Results:

  • The handheld OR-PAM system achieved an imaging resolution of approximately 7 μm.
  • Successful in vivo imaging of microvasculature in a Swiss Webster mouse ear was demonstrated.
  • The compact and flexible design significantly improves the usability of OR-PAM.

Conclusions:

  • The developed portable OR-PAM system represents a significant advancement in photoacoustic microscopy technology.
  • The system's high resolution and portability make it suitable for a wide range of potential clinical applications, particularly in dermatology.
  • This innovation enhances the accessibility and clinical utility of OR-PAM for biomedical research and diagnostics.
