When T cells with CD4 markers are activated, they give rise to two types of effector cells: helper T cells and regulatory T cells. Meanwhile, T cells with CD8 markers differentiate into effector cytotoxic T cells. Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Summary

Targeting the IL-23p19 subunit with a peptide-based vaccine (IL23-K1) significantly reduced arthritis severity in a collagen-induced arthritis (CIA) mouse model. This approach shows promise for treating autoimmune diseases like rheumatoid arthritis.

Background:

  • Interleukin-23 (IL-23) is a pro-inflammatory cytokine crucial for Th17 cell differentiation, implicated in autoimmune diseases.
  • IL-23 shares the IL-12/23p40 subunit with IL-12 and has a unique subunit, IL-23p19.
  • Previous studies showed therapeutic benefits from targeting TNF-α and IL-1β in animal models of rheumatoid arthritis.

Purpose of the Study:

  • To evaluate the efficacy of peptide-based vaccines targeting the IL-23p19 subunit in collagen-induced arthritis (CIA).

Main Methods:

  • Bioinformatic modeling identified two peptides in the murine IL-23p19 subunit.
  • Peptides were conjugated to keyhole limpet hemocyanin (KLH) to create vaccines IL23-K1 and IL23-K2.
  • DBA/1 mice were immunized with vaccines, and CIA was induced using bovine type II collagen (CIIb). Disease progression, antibody levels, cytokine mRNA, and T-cell populations were assessed.

Main Results:

  • The IL23-K1 vaccine significantly reduced clinical arthritis scores and protected against joint destruction and inflammation.
  • Mice immunized with IL23-K1 produced higher levels of anti-IL-23 antibodies compared to IL23-K2.
  • IL23-K1 immunization increased IL-10 mRNA in the spleen without affecting IL-17 levels; spleen T-cell populations remained unchanged.

Conclusions:

  • Targeting the IL-23p19 subunit via vaccination demonstrates a protective effect in collagen-induced arthritis.
  • This IL-23p19-specific vaccination strategy presents a potential therapeutic avenue for rheumatoid arthritis.
