Summary
The nucleotide sequence of plasmid-mediated beta-lactamase SHV-2 from Salmonella typhimurium was determined. It showed high similarity to Klebsiella pneumoniae LEN-1, with three amino acid differences from the E. coli SHV-2 enzyme.
Area of Science:
- Microbiology
- Molecular Biology
- Genetics
Background:
- Beta-lactamase enzymes confer antibiotic resistance.
- Plasmid-mediated beta-lactamases are significant in clinical settings.
- SHV-2 is a notable beta-lactamase found in various bacterial species.
Purpose of the Study:
- To determine the nucleotide sequence of the plasmid-mediated SHV-2 beta-lactamase (SHV-2pHT1) from Salmonella typhimurium.
- To compare the SHV-2pHT1 sequence with other known beta-lactamase genes.
Main Methods:
- Nucleotide sequencing of the SHV-2pHT1 gene.
- Bioinformatic analysis and sequence comparison.
Main Results:
- The nucleotide sequence of SHV-2pHT1 was successfully determined.
- The SHV-2pHT1 gene exhibited high similarity to the chromosomally encoded beta-lactamase LEN-1 from Klebsiella pneumoniae.
- The deduced amino acid sequence of SHV-2pHT1 differed from the Escherichia coli SHV-2 enzyme (SHV-2E.coli) by three substitutions.
Conclusions:
- Plasmid-mediated SHV-2 beta-lactamase from Salmonella typhimurium shares significant genetic similarity with other beta-lactamases.
- Amino acid substitutions may contribute to variations in beta-lactamase activity or properties.
