Myeloperoxidase (MPO) levels are elevated in Familial Hypercholesterolemia (FH) and decrease with total cholesterol (TC) reduction via LDL-apheresis (LDL-A). This suggests a link between cholesterol levels and MPO in cardiovascular disease risk.
Area of Science:
- Cardiovascular Science
- Biochemistry
- Inflammation Research
Background:
- Myeloperoxidase (MPO) is a key indicator of vulnerable atherosclerotic plaques and links inflammation to cardiovascular disease (CVD).
- Hypercholesterolemia is a known risk factor for atherosclerosis and systemic inflammation.
Purpose of the Study:
- To investigate elevated MPO serum levels in Familial Hypercholesterolemia (FH).
- To determine if acute reduction of total cholesterol (TC) impacts MPO concentration in FH patients.
Main Methods:
- Utilized LDL-apheresis (LDL-A) in FH patients to create a model of rapid TC fluctuation.
- Measured serum MPO levels via ELISA and MPO in peripheral leukocytes via immunofluorescence.
- Monitored MPO levels serially before and up to 14 days after LDL-A treatment.
Main Results:
- MPO concentrations in serum and leukocytes were significantly higher in FH patients compared to controls.
- MPO levels decreased in parallel with TC reduction during LDL-A treatment.
- Plasma TC was identified as the sole significant predictor influencing serum MPO levels (p < 0.0001).
Conclusions:
- Serum MPO levels in FH patients are directly modulated by TC concentrations, as demonstrated by LDL-A treatment.
- Further research is warranted to explore the therapeutic potential of LDL-A-induced MPO reduction in managing CVD risk.
