Summary

Absence of heat shock protein 72 (Hsp72) indicates a better prognosis in canine osteosarcoma (OSA). Widespread Hsp90 expression suggests it as a potential therapeutic target for both canine and human OSA.

Background:

  • Abnormal heat shock protein (Hsp) levels are linked to human cancers.
  • Canine osteosarcoma (OSA) serves as a valuable model for human OSA.
  • Hsps have prognostic, predictive, and therapeutic implications in neoplasms.

Purpose of the Study:

  • To investigate the expression of Hsp27, Hsp72, Hsp73, and Hsp90 in canine appendicular OSA.
  • To correlate Hsp expression with histological grade and overall survival (OS).
  • To explore the prognostic, predictive, and therapeutic potential of Hsps in canine OSA.

Main Methods:

  • Immunohistochemical analysis of 18 canine appendicular OSA samples.
  • Semiquantitative assessment of Hsp27, Hsp72, Hsp73, and Hsp90 expression.
  • Correlation of Hsp expression with histological grade and overall survival.

Main Results:

  • Hsp27, Hsp73, and Hsp90 showed variable cytoplasmic and nuclear immunoreactivity, unrelated to grade.
  • High Hsp72 immunostaining significantly correlated with grade III OSA (P < 0.01).
  • Lack of Hsp72 immunolabelling was significantly associated with longer OS (P = 0.006).
  • Hsp90 was highly expressed in all tumors and neoplastic emboli.

Conclusions:

  • Absence of Hsp72 immunosignal suggests a favorable prognosis in canine OSA.
  • Widespread Hsp90 expression indicates its potential as a therapeutic target for canine OSA.
  • Findings may inform therapeutic strategies for human osteosarcoma.
