Summary

Levodopa/benserazide treatment enhanced sensorimotor function recovery after stroke in rats by influencing astrocytes in the peri-infarct area. This study reveals potential mechanisms for dopamine-mediated stroke recovery.

Parkinson's Disease: Treatment

Neurodegenerative disorders, such as Parkinson's Disease (PD), involve the gradual and irreversible destruction of neurons in particular brain areas. These disorders exhibit standard features like proteinopathies, selective vulnerability of some neurons, and an interaction of intrinsic properties, genetics, and environmental influences in neural injury.
Parkinson's Disease is primarily a result of the loss of dopaminergic neurons in the substantia nigra pars compacta. The cornerstone of its...

Area of Science:

  • Neuroscience
  • Pharmacology
  • Stroke Research

Background:

  • Delayed levodopa/benserazide treatment in stroke patients aids functional recovery, but mechanisms remain unclear.
  • Understanding these mechanisms is crucial for optimizing post-stroke rehabilitation strategies.

Purpose of the Study:

  • To investigate if levodopa/benserazide treatment enhances neurological function recovery post-stroke in rats.
  • To explore the role of levodopa/benserazide in brain tissue reorganization after ischemic stroke.

Main Methods:

  • Rats underwent middle cerebral artery occlusion and received levodopa/benserazide or saline treatment.
  • Sensorimotor function was assessed using rotating pole, neuroscore, and cylinder tests.
  • Dopamine receptor expression and phosphoprotein levels in astrocytes were analyzed.

Locomotor Assessment of 6-Hydroxydopamine-induced Adult Zebrafish-based Parkinson's Disease Model

Published on: December 28, 2021

Main Results:

  • Levodopa/benserazide significantly improved sensorimotor function recovery without altering infarct volume.
  • Astrocytes in the peri-infarct area expressed dopamine-1 and dopamine-2 receptors.
  • Dopamine- and cAMP-regulated neuronal phosphoprotein was also found in reactive astrocytes.

Conclusions:

  • Levodopa treatment demonstrates recovery-enhancing effects post-stroke.
  • Astrocytes in the peri-infarct region may play a role in dopamine-mediated stroke recovery mechanisms.
