Ribosome Profiling02:24

Ribosome Profiling

Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.

Area of Science:

  • Cardiovascular Biology
  • Molecular Biology
  • Genomics

Background:

  • Pentraxin 3 (PTX3) is a novel vascular inflammatory marker implicated in cardiovascular health.
  • Plasma PTX3 levels rise in heart failure (HF) patients, but its cardiac expression and role in HF pathogenesis remain unclear.
  • The PTX3 gene sequence was previously unavailable for Sus scrofa (pigs), hindering cardiovascular research in this relevant animal model.

Purpose of the Study:

  • To sequence the PTX3 gene in Sus scrofa (pigs).
  • To investigate PTX3 mRNA expression in cardiac tissues of normal and HF pigs.
  • To establish a foundation for understanding PTX3's role in cardiovascular disease.

Main Methods:

  • PTX3 gene sequencing in Sus scrofa using primers designed from human sequences and Sanger sequencing.
  • PTX3 mRNA expression analysis via RT-PCR in cardiac chambers of normal and HF pigs.
  • Sequence submission to GenBank (Accession ID: GQ412351).

Main Results:

  • The Sus scrofa PTX3 mRNA sequence (1-336 bp) was successfully obtained and submitted to GenBank.
  • Pig PTX3 shared 84% sequence identity with its human homolog.
  • PTX3 mRNA was detected in all four cardiac chambers, with significantly increased expression in HF pigs compared to controls.

Conclusions:

  • The PTX3 gene sequence in pigs has been established.
  • PTX3 is expressed in the pig heart, and its expression is upregulated in a model of heart failure.
  • This foundational knowledge facilitates further research into PTX3's specific role in cardiovascular disease pathogenesis.
