Total Internal Reflection Fluorescence Microscopy01:05

Total Internal Reflection Fluorescence Microscopy

Total internal reflection fluorescence microscopy or TIRF is an advanced microscopic technique used to visualize fluorophores in samples close to a solid surface with a higher refractive index, such as a glass coverslip. TIRF only allows fluorophores in proximity to the solid surface to be excited. When light from a medium with a lower refractive index (such as air) hits the glass coverslip at a critical angle, the light undergoes total internal reflection stead of passing through the glass.

Summary
This summary is machine-generated.

Near-infrared (NIR) light can be used for medical treatments. Researchers found that neural tissue temperature rises with NIR light exposure, but this temperature increase plateaus at higher laser power levels.

Area of Science:

  • Biomedical optics
  • Neuroscience
  • Thermal imaging

Background:

  • Near-infrared (NIR) light is utilized in medical diagnostics and therapies.
  • Elevated temperatures in neural tissue are a primary safety concern during NIR light applications.

Purpose of the Study:

  • To investigate the spatial distribution of temperature changes in neural tissue under NIR light illumination.
  • To determine the relationship between laser power and temperature elevation in the brain cortex.

Main Methods:

  • An NIR light beam was applied in pulses to the brain cortex of rats.
  • Temperature mapping within the neural tissue was performed using a microprobe.

Main Results:

  • Direct measurements revealed spatial maps of temperature elevation.
  • Neural tissue temperature was observed to reach a saturation point with increasing laser power.

Conclusions:

  • The study quantifies temperature changes in neural tissue exposed to NIR light.
  • Findings suggest a limit to temperature elevation in brain tissue, irrespective of further increases in laser power, which is crucial for safe NIR medical applications.
