PDCD1 polymorphisms are not associated with Takayasu's arteritis in Turkey

Background:
- Takayasu's arteritis (TA) is a rare, chronic inflammatory condition affecting large arteries, primarily the aorta and its branches.
- The etiology of TA remains largely unknown, but associations with autoimmune diseases suggest a genetic component.
- Programmed death-1 (PD-1) protein, encoded by the PDCD1 gene, plays a crucial role in immune regulation and has been linked to various autoimmune disorders and vasculitides.
Purpose of the Study:
- To investigate the potential association between single nucleotide polymorphisms (SNPs) in the PDCD1 gene and susceptibility to Takayasu's arteritis (TA).
- To analyze PD-1.3, PD-1.5, and PD-1.6 polymorphisms within the PDCD1 gene in a Turkish population with TA.
Main Methods:
- A case-control study involving 229 TA patients meeting the 1990 ACR classification criteria and 193 healthy controls (HC).
- Genotyping of PDCD1 gene polymorphisms (PD-1.3, PD-1.5, PD-1.6) was performed using polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) analysis.
Main Results:
- No significant difference in the distribution of the PD-1.5 TT genotype was observed between TA patients and healthy controls (13.3% vs. 11.4%).
- PD-1.3 and PD-1.6 polymorphisms showed limited variability and did not differ between the study groups.
- Rare genotypes, such as PD-1.3 AA (1.4% vs. 1.0%) and PD-1.6 AG (22.4% vs. 19.2%), were similarly present in both TA patients and controls.
Conclusions:
- The studied PDCD1 gene polymorphisms (PD-1.3, PD-1.5, PD-1.6) are not associated with susceptibility to Takayasu's arteritis in the investigated Turkish population.
- These findings suggest that variations in these specific PDCD1 gene loci do not confer a predisposition to TA, despite their known links to other autoimmune conditions.
