Tight Junctions

Tight junctions are molecular seals between cells that prevent the leaking of fluids, ions, and other small solutes across cavities and compartments in multicellular organisms. They are mainly composed of claudin and occludin transmembrane proteins, and other proteins such as tricellulin and JAM (junctional adhesion molecule). Adherens Junctions

Strong contact points between adjacent cells anchor them to each other, forming tissues. Such anchoring junctions are of two types –  adherens junctions and desmosomes. Adherens junctions are abundant in tissues such as  epithelium and endothelium, forming a continuous zone of adhesion called the adhesion belt. In other tissues, such as  heart muscle, they appear as clusters, linking the cells to produce coordinated heart muscle contraction.

Summary

Nasal polyposis is linked to altered expression of tight junction proteins (TJPs). Inflammatory conditions in respiratory epithelium also reduce TJP levels, suggesting a role in persistent inflammation.

Background:

  • Tight junction proteins (TJPs) maintain epithelial barrier integrity.
  • Alterations in TJPs are implicated in various inflammatory diseases.
  • Nasal polyposis involves chronic inflammation of the sinonasal mucosa.

Purpose of the Study:

  • To investigate changes in TJP expression in nasal polyposis.
  • To examine TJP alterations in respiratory epithelium under inflammatory conditions.
  • To test the hypothesis that inflammatory cytokines alter TJP expression.

Main Methods:

  • Immunofluorescence and confocal microscopy of human sinonasal mucosa (nasal polyps vs. controls).
  • In vitro study exposing cultured human bronchial epithelium to interferon-gamma and tumor necrosis factor-alpha.
  • Western blotting to quantify claudin-1 and occludin expression.

Main Results:

  • Human nasal polyps showed reduced claudin-1 and occludin expression.
  • In vitro, cytokine exposure initially increased TJP levels but significantly reduced them after 72 hours.
  • Claudin-1 decreased by 18% and occludin by 43% after prolonged cytokine exposure.

Conclusions:

  • Nasal polyposis is associated with significant epithelial TJP alterations.
  • Inflamed respiratory mucosa exhibits similar TJP reductions.
  • These TJP changes may contribute to a self-perpetuating inflammatory state in nasal polyposis.
