Bone marrow-derived cells (BMDCs) are unlikely to initiate cancer. While BMDCs may contribute to tumor progression, they do not appear to be the source of cancer stem cells (CSCs).
Area of Science:
- Oncology
- Stem Cell Biology
- Cancer Pathogenesis
Background:
- Cancer stem cells (CSCs) are crucial for cancer development and progression.
- A key debate in cancer research concerns the origin of CSCs, specifically whether they arise from bone marrow-derived cells (BMDCs).
Purpose of the Study:
- To investigate the potential role of bone marrow-derived cells (BMDCs) in the initiation and progression of tumors.
- To determine if BMDCs contribute to the formation of cancer stem cells (CSCs).
Main Methods:
- Tumor formation was induced in female mice using n-nitrosodiethylamine (DEN) after bone marrow transplantation from male mice.
- Fluorescence in situ hybridization (FISH) and H&E staining were used to identify and quantify BMDCs in tumor and surrounding tissues.
Main Results:
- Tumor formation occurred in 20/26 mice, with various tumor types observed.
- BMDCs were found at very low frequencies within tumors (0.2-1%) and normal tissues (0-0.3%).
- A significant percentage of BMDCs (3-70%) were found in tissues surrounding tumors, correlating with inflammatory status. No evidence of fusion cells between BMDCs and tissue-specific stem cells was found.
Conclusions:
- BMDCs are unlikely to be the source of tumor initiation.
- BMDCs may play a role in tumor progression, potentially influenced by the tumor microenvironment and inflammation.
