Summary

Chitosan effectively carries the granulocyte macrophage colony stimulating factor (GM-CSF) gene, enhancing white blood cell production and cell proliferation. This suggests chitosan/pGM-CSF complexes are promising for GM-CSF therapy.

  • In vitro transfection efficiency and stability against enzymatic and serum degradation were tested in HeLa cells.

    • Main Results:

    • Full complexation was achieved at chitosan/pGM-CSF ratios of 0.1/1 and higher.
    • Complexes exhibited a size of approximately 132 nm and a zeta potential of +13.7 mV.
    • Chitosan complexes demonstrated protection against enzymatic and serum degradation, and significantly increased gene expression-dependent cell proliferation.

    Conclusions:

    • Chitosan is a suitable carrier for the pORF-hGMCSF plasmid.
    • Chitosan/pGM-CSF complexes enhance gene expression and cell proliferation, showing potential as an alternative to conventional protein treatments for GM-CSF therapy.
    • Further in vivo studies are warranted.
