Area of Science:

  • Biomedical Engineering
  • Vascular Biology
  • Animal Models

Background:

  • Peripheral arterial occlusive disease involves ischemia and impaired revascularization.
  • Gait analysis via video filming is an established model but can be subjective.
  • Objective quantification is needed to evaluate revascularization mechanisms.

Purpose of the Study:

  • To optimize a quantifiable, noninvasive, and reproducible method for analyzing ankle kinematics in rats.
  • To improve the evaluation of ischemia-induced revascularization using digital video analysis.
  • To establish a reliable computational method for gait analysis in preclinical models.

Main Methods:

  • Utilized a 2D digital video system and high-speed camera (125 fps) to film gait dynamics in hindlimb ischemic rats.

  • Developed an algorithm using Interactive Data Language (IDL) for quantitative kinematic parameter assessment.
  • Compared gait parameters between ischemic rats and sham-operated controls.
  • Main Results:

    • Stride time and knee joint angle remained significantly altered in ischemic rats 10 days post-surgery.
    • The computational analysis provided a highly reliable outline of gait kinematics.
    • Results corroborated hindlimb movement recovery linked to the revascularization process.

    Conclusions:

    • Optimized digital video analysis offers a reproducible and objective method for assessing gait in ischemic rat models.
    • This technique enhances the study of revascularization mechanisms in peripheral arterial occlusive disease.
    • The validated method supports preclinical research on vascular recovery and therapeutic interventions.
