Summary

The ayu complement C9 (aC9) protein plays a key role in fish immunity. Its expression and levels increase significantly following bacterial infection, suggesting a role in anti-bacterial defense.

Area of Science:

  • Immunology
  • Molecular Biology
  • Fish Biology

Context:

  • The complement system is crucial for innate immunity in vertebrates.
  • Complement component 9 (C9) is essential for forming the membrane attack complex (MAC).
  • Understanding fish complement components aids in aquaculture and disease management.

Purpose:

  • To clone and characterize the ayu C9 (aC9) gene and protein.
  • To investigate the expression patterns of aC9 in healthy and infected ayu fish.
  • To elucidate the role of aC9 in the immune response against bacterial pathogens.

Summary:

  • A full-length aC9 cDNA (2,125 nucleotides) encoding a 592-amino acid protein was successfully cloned.
  • aC9 mRNA expression was detected in various tissues, with the highest levels in the liver.
  • aC9 transcript levels and serum protein significantly increased post-Listonella anguillarum infection, particularly in the liver.

Impact:

  • This study identifies aC9 as a critical component of the fish immune system.
  • Findings suggest aC9's involvement in the anti-bacterial defense mechanisms of ayu fish.
  • Provides foundational knowledge for developing strategies to enhance fish disease resistance.
