Incomplete Dominance01:43

Incomplete Dominance

Gregor Mendel's work (1822 - 1884) was primarily focused on pea plants. Through his initial experiments, he determined that every gene in a diploid cell has two variants called alleles inherited from each parent. He suggested that amongst these two alleles, one allele is dominant in character and the other recessive. The combination of alleles determines the phenotype of a gene in an organism.
Trihybrid Crosses02:27

Trihybrid Crosses

Trihybrid Crosses
Some of Mendel’s crosses examined three pairs of contrasting characteristics. Such a cross is called a trihybrid cross. A trihybrid cross is a combination of three individual monohybrid crosses. For example, plant height (tall vs. short), seed shape (round vs. wrinkled), and seed color (yellow vs. green).
The F1 generation plants of a trihybrid cross are heterozygous for all three traits and produce eight gametes. Upon self-fertilization, these gametes have an equal chance to...
Epistasis Analysis01:09

Epistasis Analysis

Although Mendel chose seven unrelated traits in peas to study gene segregation, most traits involve multiple gene interactions that create a spectrum of phenotypes. When the interaction of various genes or alleles at different locations influences a phenotype, this is called epistasis. Epistasis often involves one gene masking or interfering with the expression of another (antagonistic epistasis). Epistasis often occurs when different genes are part of the same biochemical pathway. The...
Evolutionary Relationships through Genome Comparisons02:54

Evolutionary Relationships through Genome Comparisons

Genome comparison is one of the excellent ways to interpret the evolutionary relationships between organisms. The basic principle of genome comparison is that if two species share a common feature, it is likely encoded by the DNA sequence conserved between both species. The advent of genome sequencing technologies in the late 20th century enabled scientists to understand the concept of conservation of domains between species and helped them to deduce evolutionary relationships across diverse...

Summary

Using reference pedigrees with multiple relatives significantly improves the accuracy of sibling analysis. This method resolves inconclusive short tandem repeat (STR) testing results, enhancing familial relationship determination.

Area of Science:

  • Forensic genetics
  • Human genetics

Background:

  • Short tandem repeat (STR) analysis is crucial for sibship testing.
  • Inconclusive results can arise from single reference sibling comparisons.
  • Increasing STR loci offers limited improvement in problematic cases.

Purpose of the Study:

  • To evaluate the effectiveness of using multiple reference siblings in sibship analysis.
  • To demonstrate the diagnostic improvement offered by two-person reference pedigrees.

Main Methods:

  • Analysis of full sibship in paternity cases using pairs of alleged father's children.
  • Reanalysis of test siblings with low sibship likelihood against two combined reference siblings.
  • Case studies showcasing the benefits of two-person reference pedigrees.

Main Results:

  • Two-person pedigrees exponentially increased sibship probabilities for true siblings.
  • Reference pedigrees statistically verified ambiguous sibships.
  • Exclusion of alleged sibships and identification of alternative relationships were possible.

Conclusions:

  • Reference pedigrees effectively resolve ambiguous sibship questions.
  • Incorporating additional verified relatives is more powerful than increasing test loci.
  • Verified blood relatives in reference pedigrees are recommended for unclear cases.
