Antifungal Agents01:15

Antifungal Agents

Amphotericin B is a broad-spectrum antifungal agent that exploits structural differences between fungal and mammalian cell membranes. Its amphipathic structure—featuring a hydrophobic polyene-lactone ring and a hydrophilic region containing mycosamine and carboxylic acid groups—enables selective binding to ergosterol, a sterol predominantly found in fungal plasma membranes. This selective interaction underlies the drug’s antifungal activity, although weak binding to cholesterol contributes to...
Rab Cascades01:25

Rab Cascades

Rab GTPases act in a regulated cascade during membrane fusion, helping the lipid bilayers mix. The Rab family of proteins are active when bound to GTP, and inactive when bound to GDP. Hence, they act as guanine nucleotide-dependent molecular switches. Rab-GTP recognizes and binds to long or short-range tethering proteins to capture the target vesicle. These tethers coordinate with SNAREs on the vesicle and the target membrane to assemble the trans SNARE complex that locks the mixing bilayers.

Antianginal Drugs: Calcium Channel Blockers and Ranolazine

Angina pectoris, a primary symptom of ischemic heart disease, requires careful pharmacological interventions. In this context, calcium channel blockers (CCBs) and ranolazine have emerged as crucial pharmacotherapeutic agents, providing deep insights into the complexities of angina management.
CCBs, a diverse class that includes dihydropyridines (nifedipine) and diphenylalkylamines (verapamil and diltiazem), exert their effect by blocking calcium channels in cardiac and smooth muscle cells. This...

Area of Science:

  • Natural Products Chemistry
  • Mycology
  • Pharmacology

Background:

  • The higher fungal strain Coprinus radians M65 produces a variety of secondary metabolites.
  • Guanacastane skeleton-based compounds are of interest for their potential biological activities.

Purpose of the Study:

  • To isolate and characterize new diterpenoids from Coprinus radians M65.
  • To evaluate the antitumor activity of the isolated compounds.

Main Methods:

  • Isolation and structure elucidation of thirteen diterpenoids (radianspenes A-M) using chromatographic and spectroscopic techniques.
  • Antitumor activity evaluation using the MDA-MB-435 cell line.

Main Results:

  • Thirteen diterpenoids, including three lactams and one dimer, were identified.
  • Radianspene C exhibited potent inhibitory activity against MDA-MB-435 cells with an IC50 value of 0.91 μM.

Conclusions:

  • Coprinus radians M65 is a source of novel guanacastane-type diterpenoids.
  • Radianspene C shows promising antitumor potential and warrants further investigation.
