Summary
Inverse fusion PCR cloning (IFPC) offers a streamlined, PCR-based method for seamless DNA insertion into plasmids. This efficient molecular cloning technique requires minimal starting material and hands-on time, yielding numerous positive colonies.
Area of Science:
- Molecular Biology
- Gene Cloning
Background:
- Traditional cloning methods can be time-consuming and require extensive optimization.
- Seamless and directional DNA insertion into plasmids is crucial for various molecular biology applications.
Purpose of the Study:
- To introduce an easy, PCR-based three-step cloning method for seamless and directional DNA insertion.
- To enable insertion of PCR products into virtually all plasmids at a chosen site.
Main Methods:
- Inverse fusion PCR cloning (IFPC) utilizes PCR-derived inserts with vector-complementary ends.
- Overlap extension PCR facilitates fusion of insert and vector.
- Ligation and transformation are used to circularize and introduce the construct.
Main Results:
- IFPC allows for seamless and directional insertion of PCR products into plasmids.
- Minimal starting material and reduced experimental steps are required.
- High yield of positive colonies (hundreds to tens of thousands) with minimal background.
Conclusions:
- IFPC is an efficient and versatile cloning strategy.
- The method simplifies DNA cloning with reduced hands-on time.
- It is suitable for a wide range of plasmids and insertion sites.

Agarose gel electrophoresis is very useful in separating DNA fragments by size. Running a DNA ladder containing fragments of the known length alongside the sample helps determine the approximate length of the sample DNA fragments. However, additional steps are needed to verify the sequence identity of the sample DNA fragments.

RACE - Rapid Amplification of cDNA Ends
Rapid Amplification of cDNA Ends, or RACE, is one of the most effective methods to obtain a full-length cDNA from an mRNA sequence between a known internal region to the unknown sequence at the 5’ or 3’ end. The unknown region is cloned in the cDNA by a gene-specific primer that binds the known end, and a hybrid primer that attaches a predefined anchor sequence to the unknown end of the cDNA. The sequence in between is amplified by PCR with an anchor primer and a gene-specific primer.
