Area of Science:

  • Molecular Biology
  • Gene Therapy
  • Epigenetics

Background:

  • Nonviral episomal vectors are explored as alternatives to viral vectors for gene delivery.
  • Scaffold/matrix attached regions (S/MAR) enable plasmid replication and stable episomal retention in mammalian cells.

Purpose of the Study:

  • To evaluate S/MAR-based vectors as a gene therapy tool.
  • To investigate the potential of S/MAR vectors for stable genetic modification.
  • To understand the epigenetic regulation of replication and nuclear architecture using S/MAR vectors.

Main Methods:

  • Construction of expression cassettes linked to S/MAR elements.
  • Testing episomal replication and retention in various cell lines and primary cells.
  • Assessment of gene modification efficiencies compared to viral vectors.

Main Results:

  • S/MAR vectors replicate as low copy number episomes and are stably retained long-term in cells.
  • These vectors can be used for genetic modification of higher animals.
  • Gene modification efficiencies of S/MAR vectors are significantly lower than those of viral vectors.

Conclusions:

  • S/MAR vectors show potential as a stable, nonviral gene expression system.
  • Current limitations in modification efficiency hinder their application in gene therapy.
  • Technical challenges require further consideration for optimizing S/MAR vector systems.
