The absolute configuration of franganine, a cyclopeptide alkaloid from Discaria americana, was determined. This was achieved using advanced NMR spectroscopy and X-ray diffraction analysis of its salt.
Area of Science:
- Natural Product Chemistry
- Organic Chemistry
- Spectroscopy
Background:
- Cyclopeptide alkaloids are a class of natural products with diverse biological activities.
- Discaria americana is a plant source from which novel compounds can be isolated.
Purpose of the Study:
- To determine the absolute configuration of franganine, a cyclopeptide alkaloid.
- To elucidate the structure of franganine using spectroscopic and crystallographic methods.
Main Methods:
- Isolation of franganine from the methanol root bark extract of Discaria americana.
- Detailed Nuclear Magnetic Resonance (NMR) spectroscopic analysis.
- X-ray diffraction analysis of a franganine salt.
Main Results:
- The absolute configuration of franganine (1) was successfully established.
- The structure was confirmed through comprehensive NMR data.
- X-ray diffraction analysis of its salt (2) provided definitive structural information.
Conclusions:
- The absolute configuration of franganine has been unambiguously determined.
- This study provides crucial structural data for the cyclopeptide alkaloid franganine.
- The findings contribute to the understanding of natural products from Discaria americana.

Aromatic ring substitutions: Substituting the aromatic ring with –OH groups at positions 3 and 4 yields catecholamines (e.g., epinephrine), which have a high affinity for adrenoceptors. Hydrogen bonding between –OH groups and receptors enhances adrenergic activity.
