Summary
Sorting nexin 12 (SNX12) shares redundant functions with SNX3 in multivesicular endosome biogenesis. SNX12 overexpression inhibits endosome-lysosome degradation but rescues EGF receptor sorting in Hrs knockdown cells.
Area of Science:
- Cell Biology
- Molecular Biology
- Endocytic Pathway Research
Background:
- Sorting nexins (SNXs) are crucial for endosomal trafficking.
- SNX3 is known to regulate intralumenal vesicle formation in multivesicular endosomes.
- The specific role of SNX12 in the endocytic pathway remains less understood.
Purpose of the Study:
- To investigate the function of sorting nexin 12 (SNX12) in the endocytic pathway.
- To compare the functions of SNX12 with the well-characterized SNX3.
- To elucidate the role of SNX12 in multivesicular endosome biogenesis and protein sorting.
Main Methods:
- Investigated SNX12 expression levels and localization in cells.
- Analyzed the effect of SNX12 overexpression on endosomal maturation and protein trafficking.
- Examined SNX12 function in an Hrs knockdown background using EGF receptor sorting assays.
Main Results:
- SNX12 is expressed at low levels and localizes to early endosomes, dependent on 3-phosphoinositide binding.
- Overexpression of SNX12 inhibits multivesicular endosome maturation and the degradative pathway to lysosomes.
- SNX12 overexpression rescues EGF receptor sorting defects caused by Hrs knockdown.
Conclusions:
- SNX12 exhibits redundant functions with SNX3 in multivesicular endosome biogenesis.
- Despite low expression, SNX12 plays a significant role in endosomal sorting and maturation.
- SNX12 is a key regulator of the endocytic pathway, particularly in protein sorting and lysosomal degradation.

The Early Endosome: Endocytosis of Transferrin
Essential proteins such as insulin or low-density lipoprotein (LDL) and micronutrients such as iron enter a eukaryotic cell through receptor-mediated endocytosis. Subsequently, the early endosomes fuse with the vesicles containing such receptor-ligand complexes and play a vital role in sorting the incoming ligands and receptors. While the ligands are either degraded inside the vesicle or released into the cytosol, their receptors are returned to the plasma membrane for further rounds of...
SNAREs and Membrane Fusion
Once a transport vesicle has recognized its target organelle, the vesicular membrane needs to fuse with the target membrane to unload the cargo. Transmembrane proteins called SNAREs present on organelle membranes and their vesicles, mediate vesicle fusion.
SNAREs exist in pairs that symmetrically interact and catalyze the fusion of the lipid bilayers in vesicle and target organelle. v-SNARE in the vesicle membrane are single polypeptide chains that bind to a complementary t-SNARE, composed of 2...

Recycling Endosomes and Transcytosis
The recycling endosome, also known as the endosomal recycling compartment (ERC), is a part of the slow-recycling process of the endocytic pathway. Molecules internalized through receptor-mediated endocytosis are either degraded in the lysosomes or are recycled to the plasma membrane through the fast- or slow-recycling route.
The recycling endosome is not a single organelle but an extensively tubulated network of recycling pathways. It functions in storing molecules or transporting them across...

Introduction to Membrane Traffic
The ER, Golgi apparatus, endosomes, and lysosomes work in tandem to modify, sort, and package proteins and lipids. An integrated membrane trafficking network facilitates the back and forth shuttling of molecules within different organelles in the same cell or across the cell membrane.
The transport of soluble and membrane proteins is mediated by transport vesicles that collect cargo from one cellular compartment and deliver it to another by fusing with the target organelle membrane. The Rab...

Maturation of Endosomes
The early endosome containing internalized molecules matures through transformations in its location, morphology, intraluminal pH, and membrane protein composition. Together, these changes result in a more acidic late endosome that contains multiple intraluminal vesicles; therefore, the late endosome is also called a multivesicular body (MVB).
Changes in location
The maturing endosome moves along microtubules from the periphery of the cell towards the perinuclear region. This movement of the...

Clathrin Coated Vesicles
Clathrin-coated vesicles use endocytosis to transport receptors and lysosomal hydrolases from the Golgi to the lysosome in the late secretory pathway. Clathrin-mediated endocytosis was the first described endocytic process, and Clathrin-coated vesicles remain one of the most well-studied transport vesicles. The molecular machinery that generates clathrin-coated vesicles comprises over 50 proteins that precisely coordinate vesicle formation. Cell surface receptors concentrated in indented sites...
