Genetic aberrations in the Retinoblastoma gene (Rb1) were analyzed in Pakistani head and neck cancer (HNC) patients. This study identifies novel Rb1 mutations, suggesting a potential link between germline Rb1 mutations and HNC susceptibility in this population.

- Five missense mutations, two frameshift mutations, two stop codons, and two intronic substitutions were identified.
- The overall mutation frequency was 0.71.
- Novel missense mutations, a frameshift mutation, and a Lys462stop nonsense mutation were reported in Pakistani HNC patients.
Conclusions:
- Rb1 germline mutations may contribute to genetic susceptibility for HNC in the Pakistani population.
- This is the first report suggesting an association between Rb1 gene mutations and HNC risk in Pakistan.
- Further research is warranted to elucidate the role of Rb1 in HNC pathogenesis in this region.
