Summary

Phenotypic and genetic variations exist between BN, DA, and WOKW rat strains. BN rats exhibit the longest lifespan and distinct gene expression patterns in bone and adipose tissues, suggesting their utility for studying longevity genetics.

Area of Science:

  • Comparative genomics and physiology
  • Animal models in biomedical research

Background:

  • Inbred rat strains are crucial laboratory models.
  • Understanding strain-specific phenotypic and genetic variations is essential for interpreting research findings.

Purpose of the Study:

  • To investigate phenotypic and genetic differences among BN/K, DA/K, and WOKW rat strains.
  • To correlate these differences with gene expression in bone and adipose tissues.

Main Methods:

  • Studied body weight, serum constituents, and life expectancy in 95 male rats across three strains.
  • Analyzed gene expression of 20 genes in bone and adipose tissues (subcutaneous and visceral) at 30 weeks of age.

Main Results:

  • Significant differences observed in body weight, serum lipids, and leptin levels at 30 weeks.

  • BN rats demonstrated the longest life expectancy (1072±228 days) and highest bone gene expression.
  • Differential gene expression noted in adipose tissues, with specific genes showing strain- or tissue-specific patterns.
  • Conclusions:

    • Genetic variations underlie the observed phenotypic differences between BN, DA, and WOKW rat strains.
    • Gene expression studies in bone and adipose tissues support these genetic distinctions.
    • BN rats are a valuable model for investigating the genetic underpinnings of longevity.
