Summary

Interleukin-17A (IL-17A) is crucial for immunity and autoimmune diseases. Our study reveals that non-T helper 17 cells, like gamma delta T cells, are the primary producers of IL-17A during infections and autoimmune conditions.

Main Results:

  • The majority of IL-17A during K. pneumoniae infection and EAE was produced by non-CD4(+) T cells, predominantly γδ T cells, invariant NKT cells, and other CD4(-)CD3ε(+) cells.
  • IFN-γ-producing Th1 cells significantly outnumbered IL-17A-producing Th17 cells in both models.
  • IL-1β and/or IL-23 administration induced rapid IL-17A production by γδ T cells, invariant NKT cells, and other CD4(-)CD3ε(+) cells.

Conclusions:

  • Non-T helper 17 cells, particularly γδ T cells, are the major sources of IL-17A during acute immune challenges.
  • These cells are poised for rapid IL-17A production upon stimulation with cytokines like IL-1β and IL-23.
  • Findings challenge the traditional view of Th17 cells as the sole producers of IL-17A in these contexts.
