Summary

Genetic analysis of Echinococcus granulosus strains revealed distinct variations among sheep, camel, pig, and donkey isolates. Camel and donkey strains showed the closest genetic relationship, suggesting potential shared genotypic characteristics.

Background:

  • Cystic echinococcosis (CE) is a significant zoonotic disease caused by Echinococcus granulosus.
  • Understanding genetic diversity in E. granulosus is crucial for disease control and epidemiology.

Purpose of the Study:

  • To investigate genetic variations among Echinococcus granulosus cyst strains from sheep, camel, pig, and donkey.
  • To differentiate E. granulosus isolates using molecular techniques.

Main Methods:

  • Random Amplified Polymorphic DNA-PCR (RAPD-PCR) analysis was employed.
  • Seven arbitrary sequence primers were used to amplify DNA from four different host isolates.
  • Genetic similarity coefficients were calculated based on polymorphic band patterns.

Main Results:

  • RAPD-PCR generated distinct and reproducible banding patterns for each isolate.
  • Primers P1, P3, and OPH 04 effectively distinguished between the four E. granulosus strains.
  • Donkey and camel isolates exhibited the highest genetic similarity (53-78%), followed by pig and sheep (40-68%).

Conclusions:

  • Significant genetic divergence exists among Echinococcus granulosus strains from different hosts.
  • Camel isolates may share genotypic characteristics with the donkey strain, indicating potential host-strain associations.
