MicroRNAs

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns (non-coding regions of a gene) or intergenic regions (stretches of DNA present between genes). Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself, forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA...

    Summary
    This summary is machine-generated.

MicroRNA (miRNA) expression profiles can help diagnose B-cell non-Hodgkin lymphoma (NHL) subtypes. Specific miRNA signatures differentiate lymphoma types and aid in distinguishing Burkitt lymphoma (BL) from diffuse large B-cell lymphoma (DLBCL).

    Area of Science:

    • Molecular Biology
    • Oncology
    • Genetics

    Background:

    • Accurate diagnosis, prognosis, and therapy for lymphoma necessitate improved biomarkers.
    • MicroRNA (miRNA) expression is a potential area for identifying novel diagnostic markers in B-cell non-Hodgkin lymphoma (NHL).

    Purpose of the Study:

    • To investigate the relevance of miRNA expression across all major B-cell non-Hodgkin lymphoma types.
    • To identify differentially expressed miRNAs between Burkitt lymphoma (BL) and diffuse large B-cell lymphoma (DLBCL).

    Main Methods:

    • Hybridization of 147 NHL samples and 15 controls on a human miRNA platform (470 probes).
    • Comparative analysis of lymphoma types against the NHL set and direct comparison of BL vs. DLBCL.
    • Quantitative reverse transcription-polymerase chain reaction (qRT-PCR) analysis of 43 preselected miRNAs in 28 BL and 43 DLBCL samples.

    Main Results:

    • A signature of 128 miRNAs characterized lymphoma neoplasms, reflecting type, cell of origin, and oncogene alterations.
    • Comparative analysis identified 19 differentially expressed miRNAs between BL and DLBCL.
    • These miRNA differences were confirmed in a second series of 71 paraffin-embedded samples.

    Conclusions:

    • MicroRNA expression profiles offer potential as diagnostic markers for lymphoma.
    • The identified differentially expressed miRNAs are valuable for the differential diagnosis of BL and DLBCL.
