Summary

Probiotic supernatants from Lactobacillus species inhibit colon cancer cell invasion by reducing matrix metalloproteinase-9 (MMP-9) and increasing zona occludens-1 (ZO-1) protein levels. These findings suggest a potential role for probiotics in managing colorectal cancer metastasis.

Background:

  • Probiotics show promise in preventing colorectal cancer, but their role in advanced stages, particularly metastasis, is under-researched.
  • Colorectal cancer (CRC) metastasis involves complex cellular processes including invasion, influenced by matrix metalloproteinase-9 (MMP-9) and tight junction proteins like zona occludens-1 (ZO-1).

Purpose of the Study:

  • To investigate the potential of cell-free supernatants (CFS) from probiotic Lactobacillus species to inhibit colon cancer cell invasion.
  • To determine the effect of CFS on matrix metalloproteinase-9 (MMP-9) activity and zona occludens-1 (ZO-1) protein levels in metastatic colorectal cancer cells.

Main Methods:

  • Human colorectal carcinoma cells (HCT-116) were treated with CFS from Lactobacillus casei, Lactobacillus rhamnosus GG, or Bacteroides thetaiotaomicron.
  • MMP-9 activity, MMP-9 protein levels, and ZO-1 protein levels were assessed.
  • CFS were fractionated by molecular weight to identify the inhibitory components.

Main Results:

  • CFS from both L. casei and L. rhamnosus GG significantly decreased colon cancer cell invasion.
  • CFS from Lactobacillus species reduced MMP-9 protein levels and increased ZO-1 protein levels.
  • L. rhamnosus GG CFS also reduced MMP-9 activity, and inhibitory activity was found in molecular weight fractions >100 kDa and 50-100 kDa.

  • The active inhibitory factor is likely a macromolecule, suggesting potential therapeutic applications for probiotics in colorectal cancer metastasis.
