Fixation and Sectioning

Two basic types of preparation are used to visualize specimens with a light microscope: wet mounts and fixed specimens.
The simplest type of preparation is the wet mount, in which the specimen is placed in a drop of liquid on the slide. A liquid specimen can be directly deposited on the slide using a dropper. Solid specimens, such as skin scraping, can be placed on the slide before adding a drop of liquid to prepare the wet mount. Sometimes the liquid is simply water, but stains are often added...

Phase Contrast and Differential Interference Contrast Microscopy

Phase-Contrast Microscopes
In-phase-contrast microscopes, interference between light directly passing through a cell and light refracted by cellular components is used to create high-contrast, high-resolution images without staining. It is the oldest and simplest type of microscope that creates an image by altering the wavelengths of light rays passing through the specimen. Altered wavelength paths are created using an annular stop in the condenser. The annular stop produces a hollow cone of...

Area of Science:

  • Digital pathology
  • Biomedical imaging
  • Computational pathology

Background:

  • High-resolution pathology images offer extensive morphological and functional data.
  • Digital pathology imaging requires robust management of whole slide images (WSI) and derived analytical results.
  • Existing systems face challenges in managing and querying WSI data, including scalability and multi-granularity retrieval.

Purpose of the Study:

  • To develop a standard-oriented image database system (PIDB) for managing and querying digital pathology data.
  • To support integrated queries between WSI and their associated analytical results.
  • To provide a unified interface for image visualization and experiment management.

Main Methods:

  • Developed the Pathology Image Database System (PIDB) with a focus on standards compatibility and scalability.
  • Implemented a unified architecture supporting retrieval of images, tiles, regions, and analytical results.
  • Deployed PIDB for managing and querying WSI in silico brain tumor studies.

Main Results:

  • PIDB successfully manages and queries whole slide images and associated analytical data.
  • The system supports retrieval at multiple granularities (image, tile, region).
  • PIDB offers integrated querying of images and derived computational features.

Conclusions:

  • PIDB provides a generic, open-source solution for digital pathology data management.
  • The system facilitates biomedical research and clinical diagnosis through enhanced data accessibility.
  • PIDB has potential for integration with Picture Archiving and Communications Systems (PACS) for advanced pathology imaging support.
