A new copper (II) complex formulation using pullulan was optimized for treating copper deficiency. Mathematical modeling identified optimal tablet parameters for enhanced drug release and stability.
Area of Science:
- Pharmaceutical Sciences
- Materials Science
- Biochemistry
Background:
- Copper deficiency requires effective therapeutic formulations.
- Polysaccharide-based drug delivery systems offer potential advantages.
- Mathematical modeling aids in optimizing complex formulations.
Purpose of the Study:
- To develop and optimize a novel copper (II) complex formulation for treating copper deficiency.
- To investigate the impact of formulation variables on drug release characteristics.
- To establish a predictive model for tablet design.
Main Methods:
- Development of a copper (II) complex with pullulan.
- Application of mathematical modeling, including a 2(3) full factorial design.
- Utilized partial least squares (PLS) regression for optimization.
- In vitro drug release studies at pH 1.20 and 7.56.
Main Results:
- Tablet hardness significantly influenced copper (II) complex release.
- Optimal formulation (F2) achieved high drug release (98.66% at pH 1.20, 93.35% at pH 7.56).
- Formulation F2 exhibited minimal tablet weight variation.
Conclusions:
- Mathematical modeling and PLS regression effectively optimized the copper (II) complex formulation.
- The developed formulation demonstrates excellent in vitro drug release profiles.
- The approach is applicable for future copper complex formulation design.
