Overview Of Cell Separation And Isolation01:20

Overview Of Cell Separation And Isolation

Cell separation was first achieved in 1964 by S. H. Seal, who separated large tumor cells from the smaller blood cells using filtration. Two years later, Pohl and Hawk performed experiments on how cells respond differently to a nonuniform electric field based on the cell type. Such observations were the inception of cell separation methods, which allow isolating a single cell type from a heterogeneous sample.

Area of Science:

  • Cytopathology
  • Histotechnology

Background:

  • Cell block sections are crucial for cytological smear analysis.
  • Existing cell block preparation methods are continually evolving.

Purpose of the Study:

  • To introduce and evaluate a new, simple, and economical cell block preparation technique.
  • To assess the efficacy of pregelatinized starch as a cell block frame.

Main Methods:

  • A novel cell block preparation method using pregelatinized starch at room temperature.
  • Application of hematoxylin and eosin (HE) staining, immunohistochemistry, and fluorescence in situ hybridization (FISH).
  • Evaluation of 122 cytological specimens.

Main Results:

  • Pregelatinized starch effectively collects even sparse cells due to strong adhesiveness.
  • The technique is suitable for various analyses including HE, IHC, and FISH.
  • Successful application in 122 cytological specimens.

Conclusions:

  • Pregelatinized starch is a valuable and effective frame for cell block preparation.
  • This method is particularly advantageous for cytological samples with low cellularity, such as cerebrospinal fluid.
  • The technique offers a simple, economic, and efficient approach to cytological diagnostics.
