Gap Junctions01:37

Gap Junctions

Multicellular organisms employ a variety of ways for cells to communicate with each other. Gap junctions are specialized proteins that form pores between neighboring cells in animals, connecting the cytoplasm between the two, and allowing for the exchange of molecules and ions. They are found in a wide range of invertebrate and vertebrate species, mediate numerous functions including cell differentiation and development, and are associated with numerous human diseases, including cardiac and...
Gap Junctions01:27

Gap Junctions

The cytoplasm of adjacent animal cells can exchange small molecules, ions, and secondary messengers via the communication channels which form the gap junctions. These junctions comprise a few hundred to thousands of molecular channels, each made of two halves, called the connexon hemichannel. A connexon is a hexamer of six transmembrane connexin proteins, which assemble radially, thus forming a pore or channel in the center. One connexon hemichannel docks with a corresponding connexon on the...
Hair Cells01:22

Hair Cells

Hair cells are the sensory receptors of the auditory system—they transduce mechanical sound waves into electrical energy that the nervous system can understand. Hair cells are located in the organ of Corti within the cochlea of the inner ear, between the basilar and tectorial membranes. The actual sensory receptors are called inner hair cells. The outer hair cells serve other functions, such as sound amplification in the cochlea, and are not discussed in detail here.
Ligand-Gated Ion Channel Receptor: Gating Mechanism01:30

Ligand-Gated Ion Channel Receptor: Gating Mechanism

Ligand-gated ion channels are transmembrane proteins that play a vital role in intercellular communication and functions of the nervous system. They allow the influx of ions across the membrane once the neurotransmitter binds, allowing the subsequent transmission of electrical excitation across the neurons. Other ligand-gated ion channels, like the γ-aminobutyric acid (GABA) receptor, permit anions like chloride into the cells on the binding of the GABA molecule. Their entry into the cell...

Summary

Gentamicin, an aminoglycoside antibiotic, increases connexin 26 expression in rat cochlear fibrocytes. This suggests connexin 26 is involved in aminoglycoside-induced ototoxicity and acquired hearing loss.

Area of Science:

  • Ototoxicity research
  • Molecular biology of hearing
  • Drug-induced hearing loss

Background:

  • Aminoglycosides can alter ion-exchange proteins in the cochlea.
  • Connexin 26 is crucial for maintaining the endocochlear potential and endolymph potassium levels.
  • Dysregulation of connexin 26 may contribute to acquired hearing loss.

Purpose of the Study:

  • To investigate the impact of gentamicin on connexin 26 expression.
  • To elucidate the role of connexin 26 in aminoglycoside ototoxicity.

Main Methods:

  • Rats received a single dose of gentamicin.
  • Connexin 26 protein and mRNA levels were analyzed in the cochlear lateral wall.
  • Techniques included immunohistochemistry, western blotting, and real-time PCR.

Main Results:

  • Connexin 26 expression increased in type III fibrocytes post-gentamicin.
  • Elevated protein levels were observed 3 hours after gentamicin injection.
  • Increased mRNA levels were detected 24 hours after gentamicin administration.

Conclusions:

  • Connexin 26 is implicated in the acute effects of high-dose gentamicin.
  • Connexin 26 likely plays a role in the pathogenesis of ototoxic deafness.
