Summary

Brown fat inducible thioesterase variant 2 (BFIT2) is a mitochondrial protein with thioesterase activity. Its StarD14 domain binds phosphatidylinositol 4-phosphate, differentiating its function from its cytoplasmic paralog.

Area of Science:

  • Biochemistry
  • Cell Biology
  • Molecular Biology

Background:

  • Brown fat inducible thioesterase variant 2 (BFIT2), or ACOT11, is a protein with hotdog-fold and StarD14 domains.
  • Understanding BFIT2's localization and function is crucial for metabolic research.

Purpose of the Study:

  • To determine the subcellular localization and catalytic activity of human BFIT2 (hBFIT2).
  • To investigate the specific lipid-binding properties of the hBFIT2 StarD14 domain.

Main Methods:

  • HEK293 cell transfection and protein isolation.
  • In vitro thioesterase activity assays.
  • Protein-lipid overlay assays.

Main Results:

  • The N-terminus of hBFIT2 acts as a mitochondrial signal sequence, leading to its localization in the mitochondrial matrix after cleavage.

  • Mature hBFIT2 exhibits selective thioesterase activity towards long-chain fatty acyl-CoA thioesters.
  • The StarD14 domain of hBFIT2 specifically binds phosphatidylinositol 4-phosphate.
  • Conclusions:

    • hBFIT2 is a mitochondrion-localized enzyme with distinct thioesterase activity compared to its cytoplasmic paralog, CACH (ACOT12).
    • The StarD14 domain's interaction with phosphatidylinositol 4-phosphate suggests a role in specific lipid metabolism pathways within mitochondria.
