Background:

Nonsteroidal anti-inflammatory drugs (NSAIDs) are commonly used for pain and inflammation.
The combined effects of NSAIDs and exercise are not fully understood.
Exercise is often recommended for general health, but its interaction with medications requires investigation.

Purpose of the Study:

To investigate the potential negative interactions between the NSAID indomethacin (IND) and exercise in a mouse model.
To assess the impact of different doses of indomethacin combined with exercise or rest on mortality, run time, and body weight.

Main Methods:

Mice were divided into four groups: Exercise with 2.5 mg/kg IND, Sedentary with 2.5 mg/kg IND, Exercise with 5.0 mg/kg IND, and Sedentary with 5.0 mg/kg IND.

Indomethacin was administered daily via gavage one hour before a 90-minute treadmill exercise session or rest period for 14 consecutive days.

Daily monitoring included run times, body weight, and mortality rates.

Main Results:

The sedentary group receiving 5.0 mg/kg IND (Sed-5.0) exhibited high toxicity with 70% mortality.
Exercise combined with 2.5 mg/kg IND (Ex-2.5) significantly increased mortality to 52% compared to 0% in the sedentary 2.5 mg/kg IND group (Sed-2.5).
Run times were reduced from baseline in both Ex-5.0 and Ex-2.5 groups, with body weight also decreasing earlier and more significantly in the Ex-2.5 group.

Conclusions:

The combination of exercise and indomethacin use in mice can lead to severe adverse effects, including increased mortality and reduced exercise capacity.
Higher doses of indomethacin alone also pose significant toxicity risks.
Further research is warranted to explore the underlying mechanisms, such as increased gastrointestinal permeability, and to determine if similar risks exist in humans.
