Background:

  • Metastatic soft tissue sarcoma is a challenging diagnosis with limited treatment options.
  • Previous studies suggested potential efficacy of high-dose chemotherapy, but toxicity remains a concern.

Purpose of the Study:

  • To evaluate the efficacy and toxicity of high-dose cisplatin in patients with metastatic soft tissue sarcoma.
  • To determine the major response rate and safety profile of this treatment regimen.

Main Methods:

  • A Phase II clinical trial (SWOG 8465) involving 46 patients with metastatic soft tissue sarcoma.
  • Patients received high-dose cisplatin (40 mg/m2/d for 5 days), with dose adjustments for subsequent cycles.
  • Response and toxicity were assessed according to standard criteria.

  • Out of 40 evaluable patients, only six (15%) showed a partial response or no response.
  • Grade 3 or 4 leukopenia and thrombocytopenia were observed in seven and eight patients, respectively.

Conclusions:

  • High-dose cisplatin demonstrates minor activity in metastatic soft tissue sarcomas.
