Summary

Piperaquine phosphate (PQP) is not teratogenic in rats and rabbits. High doses of PQP in rats may prolong gestation and increase perinatal mortality, but do not affect lactation or pup development.

Background:

  • Piperaquine phosphate (PQP) is an antimalarial drug.
  • Assessing the safety of PQP during pregnancy and postpartum is crucial for maternal and infant health.

Purpose of the Study:

  • To evaluate the embryofetal and peri-postnatal toxicity of Piperaquine phosphate (PQP) in animal models.
  • To determine the effects of PQP on delivery, lactation, and pup development.

Main Methods:

  • Embryofetal development studies were conducted in rats and rabbits.
  • Peri-postnatal studies in rats assessed delivery, lactation, and pup growth and development.
  • Comparative analysis with other antimalarials was performed.

Main Results:

  • PQP was not teratogenic in rats and rabbits at the maximal tolerated dose.

  • Further research is warranted to elucidate the mechanism of delivery delay and to compare PQP's reproductive safety with other antimalarials like mefloquine.
