Summary

Trabectedin shows efficacy in advanced soft tissue sarcomas (STS) that have relapsed after chemotherapy. This study found it effective in unresectable and metastatic STS, particularly liposarcoma and leiomyosarcoma.

Area of Science:

  • Oncology
  • Pharmacology

Background:

  • Advanced soft tissue sarcomas (STS) have a poor prognosis, with limited survival options after standard chemotherapy.
  • Unresectable and metastatic STS progressing after anthracyclines and ifosfamide have a median overall survival (OS) of only 6 months.

Purpose of the Study:

  • To retrospectively evaluate the effectiveness and safety of trabectedin in patients with advanced, unresectable, and metastatic STS.
  • To assess trabectedin's efficacy in STS subtypes like liposarcoma and leiomyosarcoma.

Main Methods:

  • Retrospective evaluation of 8 patients with unresectable and metastatic STS.
  • Patients had relapsed or progressed after anthracycline and/or ifosfamide treatment.
  • Analysis of progression-free survival (PFS), overall survival (OS), and toxicities.

Main Results:

  • Median PFS was 3.75 months and median OS was 15 months.
  • Trabectedin demonstrated efficacy across various STS histological subtypes, including liposarcoma and leiomyosarcoma.
  • The primary toxicities observed were hematologic.

Conclusions:

  • Trabectedin is an effective treatment option for advanced STS patients who have progressed on standard chemotherapy.
  • The drug shows promise in specific STS subtypes, offering improved survival outcomes.
  • Hematologic toxicity is the main safety concern with trabectedin treatment.
