    Adenovirus-associated virus (AAV) defective parvovirus produces cytoplasmic RNA. Most AAV RNAs are spliced, with specific 5' termini and a 3' poly(A) tail at position 96.

    Background:

    • Adenovirus-associated virus (AAV) is a defective parvovirus requiring helper adenovirus for replication.
    • Understanding the structure of AAV transcripts is crucial for elucidating its gene expression and replication mechanisms.

    Purpose of the Study:

    • To characterize the structure of cytoplasmic RNA species transcribed from AAV DNA.
    • To identify the termini and splicing patterns of AAV RNAs.

    Main Methods:

    • Hybridization of AAV RNA with minus-strand DNA followed by electron microscopy.
    • Analysis of DNA.RNA duplexes using nuclease S1 or Escherichia coli exonuclease VII and agarose gel electrophoresis.

    Main Results:

    • A set of AAV RNA species was identified with 5' termini at map positions 5, 13, 19, or 39.
    • All observed RNA species possessed a 3' terminus and poly(A) tail at position 96.
    • The majority of these RNAs were spliced, lacking sequences between positions 40 and 49.
    • Unpliced RNA molecules with similar terminal positions were also detected.

    Conclusions:

    • AAV transcription generates multiple spliced and unspliced RNA species.
    • These findings provide insights into the complex RNA processing and expression strategies of defective parvoviruses.
