Purpose of the Study:

  • To investigate the role of basophils in Th17-associated inflammatory diseases.
  • To explore basophil involvement in inflammatory bowel disease (IBD) and lung inflammation.

Main Methods:

  • Detection of basophils in inflamed tissues (lung, colon).
  • Assessment of basophil influence on T cell responses, including IL-17 production.
  • Mechanistic studies involving signaling pathways (ERK1/2) and histamine receptors (H2, H4).

Main Results:

  • Basophils were found in Th17-associated lung and IBD tissues, accumulating in IL-33-rich inflamed colons.
  • Circulating basophils enhanced memory Th17 responses.
  • Activated basophils amplified IL-17 release from various CD4 T cell subsets (T(EM), T(CM), CCR6(+) CD4 T cells).
  • Basophils promoted IL-17(+)IFN-γ(-) and IL-17(+)IFN-γ(+) CD4 T cells via contact-independent mechanisms involving ERK1/2 and histamine.

Conclusions:

  • Basophils have a previously unrecognized function in augmenting Th17 and Th17/Th1 cytokine production in memory CD4 T cells.
  • Basophil accumulation in inflamed IBD tissues suggests their significance in chronic inflammatory disorders beyond Th2-associated conditions.
