Summary

A new ELISA assay accurately measures serum endothelial lipase (EL) mass. Increased EL mass correlates with cardiovascular disease and lower HDL cholesterol, suggesting a role in atherosclerosis.

Area of Science:

  • Biochemistry
  • Cardiovascular Research
  • Clinical Assays

Background:

  • Endothelial lipase (EL) is crucial for HDL cholesterol metabolism.
  • Investigating EL's role in human atherosclerosis is limited by assay availability.
  • A reliable assay for serum EL mass is needed.

Purpose of the Study:

  • To develop and validate a sensitive ELISA for quantifying serum EL mass.
  • To investigate the relationship between EL mass, HDL-C levels, and cardiovascular disease.

Main Methods:

  • Developed a sandwich ELISA using monoclonal antibodies against human recombinant EL.
  • Validated assay sensitivity, specificity, recovery, and linearity.
  • Measured serum EL mass in 645 human subjects.

Main Results:

  • The ELISA demonstrated high sensitivity (20 pg/mL) and specificity, with no cross-reactivity.
  • Serum EL mass averaged 344.4 pg/mL, with significant variation.
  • Elevated serum EL mass was observed in cardiovascular disease patients and inversely correlated with HDL-C.

Conclusions:

  • The developed ELISA is a reliable tool for measuring serum EL mass.
  • Findings support EL's role in HDL metabolism and suggest its involvement in human atherosclerosis.
  • This assay will aid further research into EL's clinical significance.
