Summary

Pazopanib significantly improved progression-free survival in patients with metastatic soft tissue sarcoma (STS). This oral medication demonstrated anti-tumor effects with an acceptable safety profile in the PALETTE trial.

  • To confirm findings from earlier phase II studies in a larger, randomized controlled trial.

Main Results:

  • Pazopanib significantly prolonged median PFS by approximately 3-fold compared to placebo (4.6 vs 1.6 months).
  • PFS benefit was observed across various tumor grades and histologies, excluding adipocytic STS.
  • Median overall survival was slightly longer with pazopanib, but the difference was not statistically significant.
  • Pazopanib did not negatively impact health-related quality of life and showed an acceptable tolerability profile.

Conclusions:

  • Oral pazopanib is an effective treatment for metastatic STS, significantly improving PFS.
  • The drug is well-tolerated and does not compromise quality of life in this patient population.
  • Pazopanib represents a valuable therapeutic option for patients with advanced STS who have progressed on standard chemotherapy.
