Enzyme-Linked Immunosorbent Assay01:33

Enzyme-Linked Immunosorbent Assay

In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane.
There are many different types of ELISAs, but they all involve an antibody molecule whose constant region binds an enzyme, leaving the variable region free to bind its specific antigen.  Enzyme-substrate reaction allows the antigen to be visualized or quantified.

Myasthenia Gravis: Diagnostic Tests

Myasthenia gravis is an autoimmune condition affecting neuromuscular transmission, causing generalized weakness in skeletal muscles. Initial diagnoses rely on patients' signs, symptoms, and medical history. The challenge lies in distinguishing myasthenia from other muscular dystrophies. An important diagnostic feature is the significant improvement of symptoms after administering anticholinesterase inhibitors.
The edrophonium test is a diagnostic tool for myasthenia gravis. It involves...

Serum Laboratory Studies, Stool Test, Breath Test

Gastrointestinal (GI) diagnostic studies are pivotal in confirming, ruling out, diagnosing, or staging various diseases, including cancers. Following diagnosis, allocating time for discussions with the patient and providing informational resources is crucial. Diagnostic assessments of the GI tract often occur in outpatient settings like endoscopy suites or GI labs. Preparation for these tests may include dietary restrictions, fasting, liquid bowel preparations, laxatives, enemas, and the...

Summary

This study developed a highly sensitive ELISA test for detecting antibodies in Aspergillosis, improving diagnosis of Aspergillus infections. A convenient diagnostic kit was also formulated and validated for clinical use.

Area of Science:

  • Medical Microbiology
  • Immunology
  • Diagnostic Development

Background:

  • Accurate diagnosis of Aspergillus-induced infections relies on detecting specific antibodies.
  • Conventional serological methods like agglutination and electrophoresis lack sensitivity and exhibit high cross-reactivity.

Purpose of the Study:

  • To characterize and identify diagnostically relevant antigens from Aspergillus fumigatus.
  • To develop a sensitive and specific diagnostic assay for Aspergillosis using these antigens.
  • To formulate a user-friendly diagnostic kit for Aspergillus antibody detection.

Main Methods:

  • Characterization and identification of Aspergillus fumigatus antigens.
  • Development of an Enzyme-Linked Immunosorbent Assay (ELISA) for detecting specific IgE and IgG antibodies.

  • Formulation of a "ready to use" diagnostic kit with dry reagents.
  • Validation of the ELISA kit using patient sera from various conditions.
  • Main Results:

    • Developed an ELISA with 92% sensitivity and 89% specificity for detecting specific IgE and IgG.
    • Successfully formulated a convenient diagnostic kit with dry reagents.
    • Validated the kit's satisfactory performance with coded sera from diverse patient groups.

    Conclusions:

    • The developed ELISA offers improved sensitivity and specificity for diagnosing Aspergillosis.
    • The formulated diagnostic kit provides a practical and effective tool for clinical use.
    • This advancement aids in the accurate diagnosis of allergic bronchopulmonary aspergillosis, aspergilloma, and invasive aspergillosis.
