We developed Fast Analysis of multidimensional NMR DAta Sets (FANDAS), a computational tool for analyzing NMR data. FANDAS aids in validating assignments, refining structures, and evaluating sample labeling using in silico methods.
Area of Science:
- Biophysics
- Structural Biology
- Computational Chemistry
Background:
- Nuclear Magnetic Resonance (NMR) spectroscopy is crucial for structural biology.
- Analyzing complex multidimensional NMR datasets can be challenging and time-consuming.
- In silico approaches can aid in the interpretation of experimental NMR data.
Purpose of the Study:
- To introduce Fast Analysis of multidimensional NMR DAta Sets (FANDAS), a computational environment for NMR data analysis.
- To enable the creation and modification of in silico NMR datasets for various analytical purposes.
- To facilitate the validation of NMR assignments and refinement of structural models.
Main Methods:
- FANDAS allows assembly of multidimensional NMR datasets from diverse input parameters.
- Input parameters include experimental NMR assignments and in silico predictions.
- Predicted datasets can be compared with experimental data using standard NMR software like Sparky.
Main Results:
- FANDAS enables rapid evaluation of sample labeling, spectral resolution, and structural content.
- The tool aids in validating NMR assignments and distinguishing molecular components.
- Demonstrated application on solid-state NMR data for a bacterial cyclic nucleotide-gated channel and sensory rhodopsin II.
Conclusions:
- FANDAS provides a versatile computational environment for advanced NMR data analysis.
- The tool supports multiple stages of NMR data analysis, from assignment validation to structural refinement.
- FANDAS is accessible as a free web portal via WeNMR.
