Regulation of Angiogenesis and Blood Supply01:24

Regulation of Angiogenesis and Blood Supply

Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. HIF1 is a dimeric protein of alpha (ɑ) and beta (β) subunits.  Under optimal oxygen conditions, HIF1β is present in the nucleus while HIF1ɑ remains in the cytosol. HIF1ɑ is hydroxylated by prolyl hydroxylase and factor...

Summary

Silent information regulator factor 2-related enzyme 1 (SIRT1) is a key enzyme regulating cellular functions. SIRT1 shows promise as a therapeutic target for diseases like erectile dysfunction linked to endothelial dysfunction.

Area of Science:

  • Biochemistry
  • Molecular Biology
  • Physiology

Background:

  • Silent information regulator factor 2-related enzyme 1 (SIRT1) is an NAD-dependent deacetylase.
  • SIRT1 regulates critical physiological functions including gene transcription, metabolism, senescence, and oxidative stress.

Purpose of the Study:

  • To explore the protective role of SIRT1 in vascular and nerve functions.
  • To evaluate SIRT1 as a potential therapeutic target for diseases involving endothelial dysfunction, such as erectile dysfunction.

Main Methods:

  • Investigated SIRT1's modulation of endothelial nitric oxide syntheses (eNOS), p53, FOXO, and NF-kappaB.
  • Assessed SIRT1's impact on endothelial and nerve protection in pathological conditions.

Main Results:

  • SIRT1 adjusts the activity of key proteins like eNOS, p53, FOXO, and NF-kappaB.
  • SIRT1 demonstrates a protective effect on vascular endothelial and nerve functions.

Conclusions:

  • SIRT1 plays a significant role in maintaining vascular and nerve health.
  • SIRT1 represents a promising therapeutic target for erectile dysfunction and related conditions characterized by endothelial dysfunction.
