Summary

We developed a whole-body gene expression atlas for Platynereis larvae, enabling high-resolution mapping of gene patterns. This method, using nuclear-stain-based registration, improves evolutionary developmental biology studies.

Area of Science:

  • Evolutionary developmental biology
  • Genomics
  • Neuroscience

Background:

  • Digital anatomical atlases are crucial for standardizing gene expression and neuronal morphology data.
  • Such atlases aid in assessing gene expression conservation across evolutionary timescales.
  • Platynereis dumerilii is ideal for these studies due to its small size and consistent development.

Purpose of the Study:

  • To develop and evaluate a whole-body gene expression pattern registration method for Platynereis larvae.
  • To create standardized reference templates for gene expression atlases.
  • To facilitate rigorous evolutionary comparisons of gene expression.

Main Methods:

  • Whole-mount in situ hybridization and confocal microscopy were employed.
  • High-resolution whole-body scanning was achieved using 2,2'-thiodiethanol (TDE) mounting medium.
  • Iterative registration and averaging of image stacks generated reference templates, with nuclear staining showing superior performance.

Main Results:

  • A method for whole-body gene expression registration in Platynereis trochophore and nectochaete larvae was established.
  • High-resolution whole-body scans were obtained using TDE, reducing aberrations.
  • Nuclear-stain-based templates enabled near-cellular-resolution gene expression registration, outperforming acetylated-tubulin-based templates.

Conclusions:

  • Whole-body reference templates for Platynereis gene expression atlases were successfully created.
  • Nuclear-staining-based image registration is a promising technique for whole-body alignment in other similar-sized organisms.
  • This work provides guidelines and scripts for expanding the Platynereis gene expression atlas.
